DNA methylation is essential for life activities and is one of the approaches of gene regulation。For instance, the methylation of CpG island located in promoter and first exon region could inhibit gene expression. Moreover, it plays an important role in tumor research, chromatin remodeling, genetic imprinting, disease research, embryonic development, and epigenetic heterogeneity so on.


Project Advice

The main idea of whole genome bisulfite sequencing (WGBS) research is about difference comparison, commonly, case vs control. After the genomic DNA was treated with bisulfite, the unmethylated C base was transformed into U base, while the methylated C base cannot be changed. After PCR amplification, the U base converts into T, which is different from the C base with methylation modification. WGBS can accurately obtain whole genome methylation profile in single base precision. It is highly recommended that 3-6 biological repeats should be set up to reduce the intra group error and enhance the reliability of the results.
